Dr. Caihong Qiu also with Yinghong Ma and Haifan Lin have published a detailed step-by-step protocol for efficiently knocking down genes in human embryonic stem cells and induced pluripotent stem cells. Advantages to this approach is that it is simple, highly reproducible, and does not require any special equipment. Using commercially available reagents (including Lipofectamine), these investigators achieved greater than 90% knock-down of key genes for maintenance of pluripotency including OCT4 and LIN28.
