Disease Models and Mechanisms Core

The Disease Models and Mechanisms Core provides services to generate unique in vivo mouse models and kidney specific cell lines.

Modification of genes of interest in bacterial artificial chromosome (BAC) for use in transgenic mice. Specific services include:

  • Selection of appropriate BACs
  • Design and verification of targeting strategies for modification
  • Performance of the modification procedure by homologous recombination in E. coli
  • Purification of BAC insert DNA ready for pronuclear injection
  • Pronuclear injection can be performed for a fee

Isolation of tubule cells and cell lines from specific nephron segments of mutant mouse strains. Specific services include:

  • Crossing mutant mice with strains to assist with immortalization or selection of tubule segments
  • Genotyping and verification of strains
  • Micro-dissection and culture of tubules
  • Selection of clonal cell lines
