Announcement of Michelle and Sam winning the CROI New Investigator award on the CROI website.

2023/11/2 Single-cell epigenetic, transcriptional, and protein profiling of latent and active HIV-1 reservoir revealed that IKZF3 promotes HIV-1 persistence” at Immunity by Yulong.

Single-cell multiomic DOGMA-seq captures in vivo states of HIV-1 DNA+ and HIV-1 RNA+ cells. HIV-1 latently infected cells are distinct at the DNA, RNA, and protein levels. HIV-1-infected cells have four distinct cell states: IRF, cytotoxic, AP-1, and cell death. We found that IKZF3 (Aiolos) promotes the proliferation of HIV-1-infected cells

Our lab visited NIH for the Strategies for an HIV Cure Meeting. Ya-Chi gave a talk on behalf of BEAT-HIV Collaboratory. Yulong and Timmy each presented a poster. Kristen presented a poster as a BEAT-HIV young investigator awardee.

2023/6/12 “Integration site-dependent HIV-1 promoter activity shapes host chromatin interaction”

at Genome Research by Jack

Using DOGMAseq (single-cell RNAseq and ATACseq), HIV-1 4C, HiC, HiChIP, CRISPRa, CRISPRi, we found that HIV-1 promoter activity increased host chromatin accessibility, increased HIV-1-host chromatin interactions in an integration site–dependent manner, within the existing chromatin boundaries. This manuscript recapitulated Jack's Uta von Schwedler Prize talk at CSHL Retro 2022. Congrats to Jack, the first and corresponding author of this manuscript!

2022/10/31 Congratulations to Dr. Jack Collora for an exceptional PhD journey!

We choose to do it not because it's easy, but because it's hard. Jack pioneered single-cell multiomics in HIV and discovered cytotoxic CD4 as a major reservoir, with exceptional curiosity, altruism, excellence, and resilience. Jack graduated in 5.5 years as a 1st author of Immunity, a co-first author of Journal of Virology, a first and corresponding author of Genome Research, and a Uta von Schwedler Prize awardee. Thanks to the committee & congrats *Dr.* Collora!

2022/6/22 “Inhibition of a Chromatin and Transcription Modulator, SLTM, Increases HIV-1 Reactivation Identified by a CRISPR Inhibition Screen”

Our CRISPRi screen found HIV silencing factors: chromatin modulators & nuclear exosome decay NEXT (ZCCHC8 & RBM7) and PAXT (ZFC3H1). SLTM knockdown in clinical samples reduces the frequency of HIV+ cells.

Mind-blowing lab retreat by glass blowing! Now we have single-particle virion made of glass (idea by undergrad Sam), fantastic beakers for maxiprep, and unique colors such as mPumpkin.

Jack gave a talk at the Cold Spring Harbor Laboratory Retroviruses meeting “The HIV-host chromatin interaction landscape” as the Uta von Schwedler Prize awardee and a poster, featured as the back cover of the meeting program book.

2022/3/22 “Single-cell multiomics reveals persistence of HIV-1 in expanded cytotoxic T cell clones”

at Immunity by Jack

Jack pioneered single-cell multiomic profiling of HIV-infected cells at their native states without ex vivo activation, for the first time in the field, and revealed that HIV persists by hiding in proliferative cytotoxic CD4+ T cell clones driven by antigen & TNF. Granzyme B inhibitor Serpin B9 may shield HIV-infected CD4+ CTLs from CD8+ CTL killing.
