Amino Acid Analysis

Amino Acid Analysis can be used to confirm the amino acid composition of peptides and proteins, determine the ratio of proteins in a complex, the % incorporation of various (non-acid-labile) modifications, the amount of contaminating protein in DNA and other non-protein samples, and which amino acids are depleted or enriched in cell media and other fluids.

The Biophysical Resource is a facility operated by Keck Foundation Biotechnology Resource Laboratory at Yale University. We currently have resources to study oligomeric state of biomolecular assemblies, as well as thermodynamics and kinetics of macromolecular interactions.

Peptide Synthesis: Large Scale

Synthetic peptides can be made routinely up to 40 residues and often, depending on sequence, up to 70 residues. All peptides that can be purified are chromatographed on a preparative C-18 or C-4 RP-HPLC system and delivered as a lyophilized material. Yields for normal peptides under 40 residues are "guaranteed" at 50 mg or more and at 90+% purity. Yields and purity are often higher, varying with the peptide sequence and length. For peptides over 40 residues or for peptides with unusual sequences, the "guaranteed" yields and purity will be considered on an individual basis.

Peptide Synthesis: Small Scale

Small scale, Fmoc peptide synthesis is carried out on a Rainin Symphony instrument that provides on instrument cleavage of the peptide from the resin. This service is available for peptides that generally range from 7 to 30 residues in length. Since the degree of difficulty in synthesizing peptides is length, composition, and sequence dependent; some peptides within this range will prove to be difficult to synthesize. Conversely, many peptides can be synthesized via this service that are longer than 30 residues.

Protein Sequencing

Amino-terminal (Edman) sequencing is carried out on an Applied Biosystems protein sequencer equipped with an on-line HPLC system. The limit of detection on this Procise 494 HT instrument is about 300 fmol of an individual phenylthio-hydantoin (Pth) amino acid. In general, 10 pmole is sufficient to sequence from 5 to 25 or more residues, with the quality of data and length of sequence that can be assigned increasing with the amount of sample. The minimum sample load is 1-2 pmole while the maximum is approximately 1 nmole. The best results are generally obtained on samples in the 10-200 pmole range.

Proteomics

The Mass Spectrometry (MS) and Proteomics Resource of the W.M. Keck Foundation Biotechnology Resource Laboratory strives to provide investigators with access to the most advanced protein profiling technologies. These technologies are implemented with a broad spectrum of mass spectrometry based techniques and protein/peptide chemistries to separate, characterize and quantify analytes from complex biological samples. The resource has 11 staff including 4 with Ph.D. level appointments, with well over 100 years of mass spectrometry and protein chemistry experience.
