Research Summary

I am interested in understanding the mechanisms of allorejection following solid organ transplantation. I am exploring the effects of both T cells and donor specific antibody on endothelial cells and evaluating the consequences of its interaction with class I vs. class II HLA molecules. To overcome the limitations of transient and incompletereduction of expression associated with RNAi in in vitro human studies, I am utilizing CRISPR/Cas9 gene editing to primary human endothelial cells, specifically by ablating expression of β2 microglobulin and CIITA, needed for class I and class II MHC expression, to create transplantable "immunoevasive" allogeneic vessels that may evade effect memory T cell recognition.
