While electron microscopy of mammalian cells has shown vesicles with approximately 70nm diameter surrounding the Golgi apparatus, conventional and confocal fluorescence microscopy do not have enough resolving power to distinguish distinct vesicular structures.

The goal of my project is to visualize vesicles near the Golgi using STORM, a super-resolution technique.

I am interested in identifying different populations of juxta-Golgi vesicles by examining the localization of COPI isoforms, Golgi resident proteins, anterograde cargo, and retrograde cargo.
